One sample prep standard, whoever is at the bench

Your intake queue changes every day. Your prep quality shouldn't.

The Singulator is the automated single-cell sample prep system for multi-sample, multi-user labs. Software-controlled protocols turn fresh, frozen, OCT, and FFPE tissue into sequencing-ready cells or nuclei, to one standard of quality, whoever is at the bench.

  • Consistent across operators and runs
  • Every sample type
  • Open reagents, your protocol
  • Automated FFPE to nuclei
  • Down to 2 mg
The Singulator 200+ automated single-cell sample prep system, with two cartridges loaded.

One instrument runs the prep your team used to do by hand

The Singulator is a benchtop instrument that automates sample prep for single-cell and single-nuclei workflows. A software-controlled protocol runs the buffer handling, timing, and temperature that a person used to run by hand, with the sample sealed inside an enclosed single-use cartridge. It takes fresh, frozen, OCT, and FFPE tissue and returns cells or nuclei ready for library prep.

Standardize your sample prep on the Singulator

A new hire's prep matches your senior tech's

A skilled manual prep already gets you the yield. The Singulator gets you that yield at a consistent quality, run to run, whoever is at the bench.

One platform for every sample you see

Your intake queue never looks the same twice: fresh, frozen, OCT, and FFPE, cells or nuclei. One walk-up Singulator runs all of it, so you take on a new sample type without a separate method for each. FFPE runs alongside fresh and frozen, and the 200+ turns a block into nuclei automatically, with no xylene and no fume hood.

Software-controlled, not software-locked

When the technician who optimized your protocol leaves, the method usually goes with them. On the Singulator, that protocol lives in the instrument, so the next hire runs it exactly as written. You can run PCS reagents or your own optimized chemistry on an open reagent platform, and every run leaves a digital record.

Handle the smallest samples

Needle biopsies, rare tissue, and limited clinical specimens are the samples you cannot afford to lose to a failed prep. The Singulator recovers nuclei from as little as 2 mg of tissue and from a 50 µm FFPE curl. For nuclei, it runs cold and enzyme-free, gentle on fragile populations.

When the standard matters most to you

Single-cell / multi-omics cores

Samples come in from every group on campus and a rotating bench team preps them. Your core's name is on the data no matter who ran the sample, so prep quality has to stay comparable across operators, shifts, and sites.

Where it fits

One software-controlled protocol runs the same way for every operator, so quality tracks the sample, not the tech at the bench.

Translational oncology

Fresh, frozen, and FFPE tumor, much of it precious: a needle biopsy you can't go back and collect again.

Where it fits

One instrument handles all three, including nuclei from as little as 2 mg.

Spatial transcriptomics

Your nuclei feed Visium HD and Xenium, where the suspension going in sets the ceiling on everything downstream.

Where it fits

Clean nuclei from the same protocol every run.

Neuroscience

Frozen and post-mortem brain for snRNA-seq, where the nuclei are fragile, and stress-sensitive populations are easily lost.

Where it fits

For nuclei it runs cold and enzyme-free to protect them.

Consistency you can check against the literature

A skilled manual prep already gets you the yield. The Singulator gets you that yield at a consistent quality, run to run, whoever is at the bench.

All performance data below is from mouse tissue. In frozen mouse cortex, Kersey et al. (2026) recorded ~100% structurally intact nuclei versus ~85% for a manual sucrose-gradient prep. Mitochondrial reads stayed under 0.5% of total reads, versus 1 to 3%. Yields matched at approximately 60,000 nuclei/mg. Ambient RNA held at rho ~0.14 on both Singulator replicates, where the manual prep ranged from 0.07 to 0.17. The study had no gentleMACS arm. In our FFPE application note (n=4, replicate sections from a single tumor block), the Singulator gave identical yields (1.0M / 1.0M) where the manual Miltenyi prep swung close to 4-fold (1.5M / 0.4M). Technical replicates produce concordant single-cell profiles, with clusters defined by biology rather than the prep method.

Where to read it yourself

One protocol, from tissue to sequencing-ready

  • Load your tissue and a single-use cartridge, pick the protocolLoad
  • The run controls buffer exchange, timing, disruption speed, and temperature on deckAutomated
  • Collect sequencing-ready cells or nucleiCollect

FFPE on the 200+: load a 50 µm curl. The instrument deparaffinizes on deck (no xylene, no fume hood). Swap to the NIC+ cartridge and collect nuclei. The 100 and 200 run the same nuclei isolation, but you deparaffinize the curl by hand first.

Why the quality holds. A manual prep rarely struggles on yield. What it struggles to do is hold that quality from one run to the next, because the same protocol still varies with the person running it. The Singulator moves those steps into a sealed, single-use cartridge and fixes the timing, disruption speed, and temperature in software, so the prep doesn't drift as it passes between people. For nuclei it runs cold and enzyme-free; cells run at 37 °C.

Where it fits. The Singulator preps the sample, a Moxi V or GO II confirms the count and viability, and you sequence on the platform you already run.

Frequently asked questions

Our senior scientist already gets excellent results. Why would we automate?

They do, and the yield data agrees. The problem shows up the week that person is away, or the month after they leave. A newer hire runs a sample you cannot repeat. The Singulator captures that expert's standard as a protocol the whole team runs the same way, so the quality holds whoever is at the bench. The senior scientist still defines the standard; the instrument makes it repeatable.

Can a single automated run match a skilled manual prep on raw yield?

A skilled manual prep already gets you the yield, and independent, peer-reviewed data found the manual prep on par with the Singulator. On some tissue a good manual prep can even edge out a single automated run. So the Singulator will not rescue a sample a good prep would lose. It gets you that yield at a consistent quality, run to run, from any operator.

Can the Singulator handle old, heavily crosslinked FFPE blocks?

Block-to-block variability is inherent to the archive, and no instrument erases it. Fixation and storage happened years before the sample reached you. The Singulator removes the operator-added variability layered on top of that: handling, timing, technique. Block selection and sectioning stay upstream. For severely degraded material, run a quick block-quality check before you commit a precious curl. PCS applications can help set the criteria.

Won't we get locked into your protocols and reagents?

No. The Singulator is an open reagent platform. Run PCS's tissue-specific enzymes, or bring the chemistry you have already optimized and run it on the instrument. The validated protocol is saved in the instrument's library, ready to run or update. On consumables, the number that matters is cost per usable sample rather than cartridge cost per run. One avoided re-run pays for many cartridges.

Doesn't any automated disruption introduce stress-response artifacts?

Any method that breaks tissue apart, by hand or automated, can trigger stress pathways. No prep is free of it. Warm enzymatic dissociation at 37 °C in particular can induce stress-response and microglial-activation signatures (Marsh et al. 2022). For nuclei the Singulator runs cold and the run is short, which limits the stress. Evaluate the data against your own internal controls.

Will ultra-low input mean low yield or compromised quality?

Low input is exactly where gentle chemistry earns its place. On the nuclei side, cold, enzyme-free processing and short run times protect the material you have rather than grinding it away. Nuclei recovery is validated at inputs as low as 2 mg. For any new low-input tissue, run a quick quality-control (QC) check against your own baseline. PCS applications will support the first samples.
